Spielberg and his wife, Kate Capshaw, have donated $100,000 to fight the initiative, called Proposition 8, they announced in a statement Monday.
"By writing discrimination into our state constitution, Proposition 8 seeks to eliminate the right of each and every citizen in our state to marry regardless of sexual orientation," the statement said. "Such discrimination has no place in California’s constitution, or any other."
Also known as the Marriage Protection Act, Proposition 8 would amend the state constitution to limit marriage to a man and a woman. If passed, it would overturn the state Supreme Court decision that made the state only the second in the United States to legalize same-sex marriage.
Actor Brad Pitt gave the same amount to the cause last week.

The You, Me and Dupree star says she likes to rehearse sex scenes so she knows what she is doing and has something to concentrate on before filming starts.
She explains, "I like to prepare for love scenes with lots of rehearsal. But in reality you just get on with it. You have that moment of 'OK here we go' then you just have to take a deep breath and get on with it."
Kate, 29, also makes sure she has fresh breath before filming intimate scenes.
She added, "I make sure I've had my fresh breath mints.
"At the time of filming My Best Friend's Girl I was smoking, so that must have been nice for Dane Cook. But I've quit now."

Actress Lindsay Lohan has become the center of a political tug-of-war between presidential hopefuls John McCain and Barack Obama.
Obama reportedly rejected an offer from Lohan -- who, by her own admission, is a reformed drug and alcohol addict -- to host a campaign fundraiser in an effort to rally support for his candidacy among young voters.
A source tells the Chicago-Sun Times the Democrat's camp declined Lohan's offer because she "is not exactly the kind of high-profile star who would be a positive for us."
McCain has challenged his opponent's judgment, with a spokesperson telling celebrity Web site TMZ.com: "So let me get this straight -- they turned away Lindsay Lohan, but Barack Obama has friends like unrepentant terrorist Bill Ayers and convicted felon Tony Rezko? Maybe LiLo is just too upstanding for Barack Obama."

Rihanna is set to be the latest celebrity to dabble in the world of fashion, with the news that she will launch her own clothing range.
In Milan yesterday for the Gucci show, the Disturbia singer told WWD that the collection was already in the works.
'I'm not one to rush into anything. I want to take my time,' she said, I can’t say when it will be released, but it's definitely going to happen.'
As the latest spokesperson for Gucci, Rihanna shot her first campaign for the brand over the summer. When asked why she joined the brand she responded that its charity work had been a deciding factor, 'I give them so much respect,' she described.

Paparazzi Alison Silva had claimed in a lawsuit that Reeves intentionally drove into him while behind the wheel of his Porsche 911 in March 2007, causing injuries and a loss of earnings for the photographer.
However Los Angeles Superior Court judge Elizabeth Grimes said there were no facts to support Silva's claims of assault against "Matrix" star Reeves.
"There was no evidence of the slightest contact or the slightest touching," Grimes said. "It didn't happen. Plainly Mr. Reeves intended to drive away. That intent is not unlawful."
The judge said Silva had fallen when he tripped on his own feet, adding that Reeves himself appeared to have been the victim of an assault by Silva when the photographer put his hands on the actor's car.
Reeves, who was not in court for Monday's ruling, said in a sworn statement submitted in July that he did not try to harm the photographer.
"At no time did my vehicle make contact with Mr. Silva other than when (he) bent over and placed his hand on the hood of my car," Reeves' statement said.
Silva will continue to sue Reeves for negligence in connection with the incident in Rancho Palos Verdes, a posh neighborhood in southwest Los Angeles.
